We have got a result on the comparison of polyscaffold and basic scaffold. However, it did not turn out to be what we expected. As can be seen in the following graph, although the fluorescence of polyscaffold at 20℃ is more intense than that of basic scaffold at 37℃, the intensities of fluorescence of polyscaffold are about 40% lower than those of basic scaffold at both 20 and 37℃.
To explain the result, we raise two possible reasons. The first one is that the added sequence decreases the stability of basic scaffold, which contribute to the degradation of the monoscaffolds. The second is that the linear polyscaffolds fold into clews, thus leading to the binding domain on MS2 and PP7 aptamers being hided inside the clews.
